Electrochemical Detection of in Situ Oxidative Damage to Dna

چکیده
Electrochemical voltammetric in situ detection of DNA oxidative damage caused by reduced adriamycin, an antibiotic of the family of anthracyclines, intercalated into DNA was possible using a DNA-electrochemical biosensor. The results indicate that adriamycin intercalated in double helix DNA can undergo oxidation or reduction and cause oxidative damage to DNA by reacting specifically with the guanine moiety in guaninerich regions at CpG homologous sequences, and thence to the formation of mutagenic 8-oxoG residues, the main product of guanine oxidation.

Triazole-linked phenyl derivatives: redox mechanisms and in situ electrochemical evaluation of interaction with dsDNA.
The redox mechanism of two trisubstituted triazole-linked phenyl derivatives (CL41 and CL42) and a disubstituted triazole-linked phenyl derivative (CL2r50) were studied using cyclic, differential pulse and square wave voltammetry at a glassy carbon electrode. The CL41, CL42 and CL2r50 oxidation is a complex, pH-dependent irreversible process involving the formation of electroactive products tha...

Genetically modified organisms (GMOs) are plants or animals whose genetic make-up has been transformed by recombinant DNA technology, which has new features such as resistance to herbicides, virus and insect. Recently, genetic modification of food products has increased in order to reduce world poverty and hunger and increase food production However, the impact of GMOs on the human health is a ...
متن کاملIn situ evaluation of heavy metal-DNA interactions using an electrochemical DNA biosensor.
Heavy metal ions, lead, cadmium and nickel, are well known carcinogens with natural different origins and their direct mode of action is still not fully understood. A dsDNA-electrochemical biosensor, employing differential pulse voltammetry, was used for the in situ evaluation of Pb2+, Cd2+ and Ni2+ interaction with dsDNA. The results confirm that Pb2+, Cd2+ and Ni2+ bind to dsDNA, and that thi...
متن کاملOGG1 DNA Repair Gene Polymorphism As a Biomarker of Oxidative and Genotoxic DNA Damage
Background: Single nucleotide polymorphisms in 8-oxoguanine DNA glycosylase-1 (OGG1) gene modulates DNA repair capacity and functions as one of the first lines of protective mechanisms against 8-hydroxy-2’-deoxyguanosine (8-OHdG) mutagenicity. OGG1-Cys326 gene polymorphism may decrease DNA repair function, causing oxidative stress due to higher oxidative DNA damage. The main purpose of this stu...

Objective Titanium dioxide (TiO2) is a white pigment which is used in paints, plastics, etc. It is reported to induce oxidative stress and DNA damage. The N-acetylcysteine (NAC) was used to fight oxidative stress-induced damage in various tissues. The aim of this study was to evaluate the toxic effects of TiO2 nanoparticles by oral administration and the protective role of NAC on testes of a...
